Abstract:
One hundred and sixty isolates of mold and forty isolates of bacteria capable of using chitin as carbon source were isolated from various soil samples, lignocellulosic waste , decayed plant materials and stock culture of the Microbiology Laboratory, Faculty of Science, Chiang Mai university . The isolates were tested for the production of chitinase in enzyme production medium (EPM) over 7 days for mold and 5 days for bacteria. The highest chitinase activity was obtained from the mold isolate B11 isolated from soil at Doi Suthep forest. This strain was identified as Geotrichum sp. The yield of chitinase was 100.0 milliunit (mU) /ml giving the specific activity of 588.0 mU/mg. The isolate grew most rapidly on malt extract glucose agar (MYG) medium, pH 4.5-9 at room temperature (28± 2° C). Investigation of the conditions for the chitinase production by Geotrichum B11 showed that the inoculum size of 8% in a liquid medium pH 5.5, containing 0.5% colloidal chitin as carbon source and (NH4)2SO4 as nitrogen source gave the highest yield of the enzyme at 37° C. The maximum enzyme activity was obtained after 3 days reaching 225 mU/ml and specific activity of 198.0 mU/mg.
